RESUMO

During diabetes, the characteristic hyperglycemia can induce red blood cell glycation. Several researchers have proposed different protocols to perform an in vitro model to study this phenomenon. In this article, some of the most important in vitro glycation protocols available in the bibliography were compared to each other. The incubation parameters as the suspension medium, glucose concentration, red blood cell concentration, time, and temperature were analyzed. Also, several assays were carried out in our laboratory, and glycated hemoglobin, erythrocyte aggregation and viscoelasticity were determined for the protocol validation. Based on the bibliographic analysis and our experimental results, an optimal protocol for in vitro glycation of red blood cells is presented.

Several studies report flow disturbance and microcirculation disorders upon anesthesia treatment. These alterations are often related to blood rheology changes. In this work, it was attempted to make a detailed description of the alterations in erythrocyte mechanical properties by the action of propofol, remifentanil, and vecuronium. For this, an in vitro study was performed on red blood cell samples from healthy donors incubated with solutions of propofol (4 µg/mL whole blood), remifentanil (10 ng/mL plasma), and vecuronium (0.15 µg/mL plasma). Erythrocyte viscoelastic parameters were determined by octuplicate using a Reómetro Eritrocitario. Also, a Wilcoxon signed rank-test with Yates correction for continuity was performed to analyze the overall alteration in the mechanical properties of erythrocytes. Statistical analysis showed that the three studied anesthetics changed the erythrocyte mechanical properties at different parts of the membrane. These results would imply an interaction of these anesthetics with the erythrocyte membrane. Finally, this could conduce to alterations in microcirculation.

The simultaneous determination of adhesion and deformability parameters of erythrocytes was carried out through a microfluidic device, which uses an inverted optical microscope with new image acquisition and analysis technologies. Also, an update of the models describing erythrocyte adhesion and deformation was proposed. Measurements were carried out with red blood cells suspended in saline solution with human serum albumin at different concentrations. Erythrocytes adhered to a glass surface were subjected to different low shear stress (from 0.04 to 0.25 Pa), causing cellular deformation and dissociation. The maximum value obtained of the erythrocyte deformability index was 0.3, and that of the adhesion energy per unit area was 1.1 × 10-6 Pa m, both according to previous works. The obtained images of RBCs adhered to glass reveal that the adhesion is stronger in a single point of the cell, suggesting a ligand migration that concentrates the adhesion in a "spike-like tip" in the cell. Moreover, adhesion energy results indicate that the energy required to separate erythrocytes in media with a lower albumin concentration is greater. Both results could be explained by the mobility of membrane receptors.

INTRODUCCIÓN: las emulsiones son un tipo de preparado farmacéutico muy utilizado en aplicación tópica consistentes en sistemas bifásicos de aceite-agua o agua-aceite, donde el principio activo deseado se incorpora en una de las fases dependiendo de su solubilidad. Diversos estudios demuestran que la estabilidad es mayor en emulsiones con estructuras líquido-cristalinas. El principio activo liposoluble Miconazol, que actúa como antimicótico, se solubiliza en la fase oleosa de la emulsión y en la fracción de la cadena hidrocarbonada de los cristales líquidos. MÉTODO: se utilizaron técnicas microscópicas para analizar las características de una emulsión convencional y de otra con cristales líquidos, a las que se les incorporó el principio activo Miconazol. Se determinaron las dimensiones de las gotas de la fase interna y mediante microscopía de polarización se caracterizaron los cristales líquidos. RESULTADOS: el análisis de las imágenes microscópicas permitió determinar que en las formulaciones con cristales líquidos con y sin Miconazol, aproximadamente el 80 % de las gotas tienen dimensiones en el intervalo 0,5 mim - 1 mim. Las observaciones microscópicas con luz polarizada nos permitieron determinar que los cristales líquidos tienen birrefringencia con la formación de cruces de extinción uniáxicas negativas, las cuales son características de las fases liotrópicas laminares con texturas cónicas focales. CONCLUSIONES: los resultados muestran que el agregado de Miconazol, no interfiere con la formación de la estructura de los cristales líquidos, por lo que estas dependen de los componentes de la formulación y de la técnica de preparación


INTRODUCTION: emulsions are a type of pharmaceutical preparation widely used in topical applications consisting of two-phase systems of oil-in-water or water-in-oil, where the desired active ingredient is incorporated into one of the phases depending on its solubility. Several studies show that stability is greater in emulsions with liquid-crystalline structures. The liposoluble active substance Miconazole, which acts as an antifungal agent, is solubilized in the oil phase of the emulsion as well as in the fraction of the hydrocarbon chain in liquid crystals. METHOD: microscopic techniques were used to analyze the characteristics of both a conventional emulsion and another one containing the liquid crystals. Miconazole was incorporated into both emulsions; drop dimensions in the internal phase were determined and the liquid crystals were characterized by polarization microscopy. RESULTS: through the analysis of the microscopic images of the formulation with liquid crystals with Miconazole and without Miconazole, it was possible to determine that approximately 80% of the drops have dimensions ranging from 0.5 Mum - 1 Mum. Microscopic observations with polarized light allowed us to determine that liquid crystals have birefringence with the formation of negative uniaxial extinction crosses, which are characteristic of laminar lyotropic phases with focal conical textures. CONCLUSIONS: the results show that the addition of Miconazole does not interfere with the formation of the structure of the liquid crystals. Therefore, the formation of liquid crystals depends both on the components of the formulation and the preparation technique

BACKGROUND: A highly reduced expression of Rh antigens in the erythrocyte membrane is the main feature of Rhmod , an extremely rare phenotype. Mutations within RHAG gene, which encodes RhAG glycoprotein and modulates Rh antigen expression and Rh complex formation, are the molecular events responsible for the Rhmod phenotype. Here we report a clinical, serologic, and molecular study of an Argentinean proband with Rh-deficiency syndrome. MATERIALS AND METHODS: Rh antigens, RhAG and CD47 glycoproteins were studied by serologic methods in the proband, her parents and sister. Osmotic fragility and viscoelastic parameters were also examined. RHD zygosity was analyzed by RFLP-PCR. RHD, RHCE, and RHAG genes were studied by Sanger sequencing. RESULTS: No Rh antigens were detected in the proband by standard techniques. However, adsorption-elution and anti-RhAG tests showed that the proposita was Rhmod . Reduced expression of CD47, enhanced osmotic fragility, and surface viscosity alterations giving rise to spherocytes were observed in the patient. Sequencing analysis showed that a c.920C>T mutation in RHAG Exon 6 was present in a homozygous state in the proband and in a heterozygous state in the rest of the family. This novel missense mutation caused the p.Ser307Phe amino acid substitution in Transmembrane Segment 10 of the RhAG glycoprotein. CONCLUSION: This comprehensive study determined the causes of the proband's anemia allowing the diagnosis of Rh-deficiency syndrome.

Objetivos: La diabetes mellitus es señalada como una de las principales amenazas para la salud humana en el siglo XXI. Entre las plantas medicinales utilizadas tradicionalmente para su tratamiento, se encuentra la Bauhinia forficata que crece en Argentina y Brasil. En este trabajo se realizó un estudio comparativo de parámetros reológicos y fisicoquímicos entre diferentes extractos acuosos obtenidos a partir de hojas de Bauhinia forficata recolectadas en la ciudad de Rosario (Argentina) y hojas comerciales vendidas en dietéticas y farmacias. Métodos: Se prepararon soluciones extractivas a partir de las hojas disecadas y solución fisiológica, utilizando los siguientes procedimientos: Cocimiento, Digestión Controlada, Infusión y Maceración. Se realizó la caracterización reológica, óptica y fisicoquímica de los mismos. Resultados y Conclusiones: Los resultados obtenidos son de utilidad para el análisis de la hemocompatibilidad de estos extractos, contribuyendo además a los estudios para la comprensión de sus mecanismos de acción como antidiabéticos en fitomedicina


Objectives: Diabetes mellitus is identified as one of the main threats to human health in the 21st century. Among the medicinal plants used traditionally for its treatment, is the Bauhinia forficata that grows in Argentina and Brazil. This work makes a comparative study of rheological and physicochemical parameters between different aqueous extracts, obtained from leaves of Bauhinia forficata collected in the city of Rosario (Argentina) and commercial leaves sold in diet and pharmacies. Methods: Extractive solutions were prepared from the dissected leaves and physiological solution, using the following procedures: Cooking, Controlled Digestion, Infusion and Maceration. The rheological, optical and physicochemical characterization was carried out. Results and Conclusions: The results obtained are useful for the analysis of the hemocompatibility of these extracts, contributing also to the studies for the understanding of their mechanisms of action as antidiabetics in phytomedicine

The study of red blood cell (RBC) aggregation is of great interest because of its implications for human health. Altered RBC aggregation can lead to microcirculatory problems as in vascular pathologies, such as hypertension and diabetes, due to a decrease in the erythrocyte surface electric charge and an increase in the ligands present in plasma. The process of erythrocyte aggregation was studied in stasis situation (free shear stresses), using an optical chip based on the laser transmission technique. Kinetic curves of erythrocyte aggregation under different conditions were obtained, allowing evaluation and characterization of this process. Two main characteristics of blood that influence erythrocyte aggregation were analyzed: the erythrocyte surface anionic charge (EAC) after digestion with the enzyme trypsin and plasmatic protein concentration in suspension medium using plasma dissolutions in physiological saline with human albumin. A theoretical approach was evaluated to obtain aggregation and disaggregation ratios by syllectograms data fitting. Sensible parameters ( Amp 100 , t 1 \ 2 ) regarding a reduced erythrocyte EAC were determined, and other parameters (AI, M-Index) resulted that are representative of a variation in the plasmatic protein content of the suspension medium. These results are very useful for further applications in biomedicine.

Drugs used during anesthesia might induce disturbance on microcirculation due to their systemic cardiovascular actions and to direct hemorheological effects. A comparative investigation of the hemorheological alterations related to in vitro propofol treatment of red blood cells (RBCs) from healthy and diabetic volunteers is presented here. Viscoelasticity and aggregation of RBCs from type 2 diabetic patients (DBT) and healthy donors (HD) were studied from RBCs incubated with propofol near steady-state concentration. 'S parameter', which measures the aggregation degree, was obtained using digital analysis of microscopic images. Erythrocyte viscoelasticity parameters were determined using an Erythrocyte Rheometer. Results obtained from DBT samples showed an increase of 10% or more in aggregation due to the propofol action. The phase shift between erythrocyte response and oscillating shear stress applied at 1 Hz was altered by propofol treatment of erythrocyte from HD and DBT. Propofol could produce slight alterations in the rheological behavior of erythrocyte from HD and DBT, at concentrations near those of steady state. Moreover, this anesthetic could induce an adverse effect in DBT, particularly on erythrocyte aggregation. The observed hemorheologic alteration would increase the possibility of microcapillary obstruction. Hence, this type of study [0] would prove relevant to avoid possible postoperative complications.

Phyllanthus sellowianus extracts have been used in Argentina since colonial times in the treatment of diabetes. The in vitro biorheological and hemoagglutinant action of different extracts of P. sellowianus bark on human erythrocytes (RBC) were studied. RBCs were incubated in vitro with four aqueous extracts: Maceration; Controlled Digestion (PD); Decoction; and Infusion. Biorheological parameters (deformability, membrane surface viscosity, elastic modulus, and dynamic viscolelasticity) were determined with an Erythrodeformeter, and erythrocyte adhesion was characterized by image digital analysis. Immunohematological assays in RBC incubated with all the extracts showed large globular aggregates and agglutination in human ABO blood group system. Isolated cell coefficient showed the increase of cell adhesion. Aggregated shape parameters were significantly higher than normal and they changed with the concentration, particularly of PD extracts. Rheological results showed that the extract biorheological action varies with the temperature used in the extract preparations. The results obtained are useful to study the action mechanism of extracts from P. sellowianus bark in order to evaluate its use as therapeutic agent in diabetes. Immunohematological Tests using ABO system showed its agglutinant power, which is of special interest in Immunohematology to be used as hemoclassifier.

Previous studies have shown that A. lumbricoides extracts capture sialic acid (SA) from human red blood cells (RBC). The aim of this work was to study hemorheological alterations in vitro caused by parasite larvae. The biorheological action of three larva concentrates of first and second larval stage on group O erythrocytes was analyzed by incubating the erythrocyte packed together with an equal volume of larvae (treated RBC) and PBS (control RBC). Distribution and parameters of aggregation (digital image analysis), aggregation kinetics (erythroaggregameter), and viscoelasticity (erythrodeformeter) were measured. The digital image analysis showed that all the larvae diminished the isolated cells percentage and increased the size of the formed aggregates. The aggregate formation velocity was lower in the treated than in the control. The deformability index (ID) values of treated RBC did not present variations with respect to those of the control, but a decrease in the erythrocyte elastic modulus (µ(m)) and membrane surface viscosity (η(m)) values was observed, indicating that the larvae not only induced a diminution in the membrane surface viscosity of RBC but also altered the dynamic viscoelasticity of the membrane. Experiments carried out in vitro support the conclusion that the contact between larvae and RBC produces hemorheological alterations.

Experiencias previas han reportado que los extractos de A. lumbricoides captan ácido siálico del eritrocito humano modificando su carga. El objetivo fue evaluar in vitro las posibles alteraciones biorreológicas debidas al efecto de extractos del parásito adulto. Se analizó la acción de 8 extractos sobre eritrocitos Grupo O, incubando el sedimento globular con igual volumen de extracto y con PBS. Se determinó la distribución y morfología de los agregados (Análisis Digital de Imágenes), carga porcentual (método Azul de Alcian), cinética de agregación (eritroagregámetro) y viscoelasticidad (eritrodefórmetro). Los resultados mostraron que todos los extractos causaron una disminución del porcentaje de células aisladas y un aumento en el tamaño de los agregados, sin alterar su morfología. Los glóbulos tratados con 6 extractos mediaron una significativa alteración de la carga superficial y con 5 de ellos la velocidad de agregación fue menor que en el control. Se observó una correlación lineal entre los parámetros de cinética y la concentración proteica de los extractos. Uno de los tratamientos modificó la fluidez de membrana y dos disminuyeron la deformabilidad eritrocitaria, pero ninguno afectó la elasticidad. Estas experiencias in vitro permiten concluir que el contacto de los extractos con los eritrocitos produce alteraciones hemorreológicas.


Previous studies have reported that A. lumbricoides extracts capture sialic acid from human erythrocytes, modifying their charge. The purpose was to evaluate in vitro the possible biorheological alterations due to the effect of extracts from adult parasites. The activity of 8 extracts over Group O erythrocytes was analyzed incubating the globular sediment with an equal volume of extract and with PBS. The distribution and morphology of the aggregates was determined (Digital Image Analysis), percent of charge (Alcian Blue method), aggregation kinetics (erythroaggregameter), and visco-elasticity (erythroderformeter). The results showed that all the extracts produce a decrease of the percentage of isolated cells and an increase of the size of aggregates, without altering their morphology. Red cells treated with 6 extracts mediated a significant alteration of their superficial charge and with 5 of them the aggregation velocity was less than that of the control. A lineal correlation between the kinetic parameters and the protein concentration of the extracts occurred. One of the treatments modified the fluidity of the membrane and two decreased erythrocyte deformability, but none of them affected elasticity. These in vitro experiences allowed concluding that contact of the extracts with the erythrocytes produces hemorheological alterations.

La caracterización de agregados eritrocitarios es importante para analizar las posibles alteraciones en la microcirculación observadas en ciertas patologías vasculares como la hipertensión y la diabetes. El objetivo de este trabajo fue estandarizar una técnica que pueda ser utilizada por cualquier operador con un equipamiento similar. Para ello se prepararon distintas suspensiones de glóbulos rojos de dadores sanos en plasma autólogo, que fueron observadas con un microscopio óptico invertido. Se registraron para su análisis las imágenes de los agregados con una cámara digital. Se realizaron los recuentos de células individuales, agregados de 2 a 4 células, agregados de 5 ó más células y amas (redes de agregados de gran tamaño). Se midió el perímetro y el área, obteniéndose un parámetro de forma (ASP) de cada agregado de 5 ó más células. Los resultados obtenidos permitieron estandarizar el protocolo de trabajo concluyendo que la dilución óptima de glóbulos rojos en plasma autólogo para esta técnica es 0,5%.


Characterization of erythrocyte aggregates is important in the analysis of the possible alterations observed in the microcirculation of certain vascular pathologies such as hypertension and diabetes. The objective of this work was to standardize a technique that can be used by any operator having similar equipment. For that purpose, different suspensions of red blood cells from healthy donors were prepared in autologous plasma and then observed with an inverted light microscope. The images of the aggregates were recorded with a digital camera in order to be later analyzed. Individual cell count was carried out, as well as 2 to 4 cell- aggregates, 5 or more cell- aggregate and amas (big aggregate networks). Measurement of perimeter and area of each of the aggregates made up of 5 or more cells was performed, getting a shape parameter (ASP). Due to the results obtained, this working protocol has been standardized and it can be concluded that the optimal dilution of red blood cells in autologous plasma is 0.5% for this particular technique.


A caracterizagáo de agregados eritrocitários é importante para analisar as possíveis alteragóes na microcirculagáo observadas em certas patologias vasculares como a hipertensáo e a diabetes. O objetivo deste trabalho foi padronizar uma técnica que possa ser utilizada por qualquer operador com um equipamento similar. Para isso foram preparadas diversas suspensóes de glóbulos vermelhos de doadores saudáveis em plasma autólogo, que foram observadas com um microscópio óptico invertido. Foram registradas para a sua análise as imagens dos agregados com uma camera digital. Realizaramse as recontagens de células individuais, agregados de 2 a 4 células, agregados de 5 ou mais células e amas (redes de agregados de grande tamanho). Foi medido o perímetro e a área, obtendo um parametro de forma (ASP) de cada agregado de 5 ou mais células. Os resultados obtidos permitiram padronizar o protocolo de trabalho concluindo que a diluigáo ótima de glóbulos vermelhos em plasma autólogo para esta técnica é de 0,5%.

La caracterización de agregados eritrocitarios es importante para analizar las posibles alteraciones en la microcirculación observadas en ciertas patologías vasculares como la hipertensión y la diabetes. El objetivo de este trabajo fue estandarizar una técnica que pueda ser utilizada por cualquier operador con un equipamiento similar. Para ello se prepararon distintas suspensiones de glóbulos rojos de dadores sanos en plasma autólogo, que fueron observadas con un microscopio óptico invertido. Se registraron para su análisis las imágenes de los agregados con una cámara digital. Se realizaron los recuentos de células individuales, agregados de 2 a 4 células, agregados de 5 ó más células y amas (redes de agregados de gran tamaño). Se midió el perímetro y el área, obteniéndose un parámetro de forma (ASP) de cada agregado de 5 ó más células. Los resultados obtenidos permitieron estandarizar el protocolo de trabajo concluyendo que la dilución óptima de glóbulos rojos en plasma autólogo para esta técnica es 0,5%.(AU)

Measurement of perimeter and area of each of the aggregates made up of 5 or more cells was performed, getting a shape parameter (ASP). To mask the antigenic sites of cells for cell therapies, especially for blood transfusion, we investigated the hemocompatibility of two poly(2-(dimethylamino)ethyl methacrylate-co-poly(ethyleneglycol) compared with that of the homopolymer without PEG. Our strategy relies on the potential ability of these copolymers to self-assemble at the erythrocyte surface. The cationic sequence of the copolymer should be able to interact with the glycocalyx by ionic interaction. The other sequence, based on a polyethyleneglycol moiety, should prevent both nonspecific interactions and specific recognition of the biological surface. The hemocompatibility of these copolymers was assessed by analyzing alterations in human erythrocyte membrane viscoelasticity, morphology, granularity, and aggregation. Their properties to mask ABO system and three erythrocyte glycophorin sites were investigated. No alterations in the erythrocyte morphology were observed by confocal microscopy. On the other hand, a partial masking of different specific glycophorin sites leads to future optimization of the macromolecular structures of these functionalized copolymers.

The aim of this study was to analyze the strength of red blood cells agglutination, induced by autoantibodies in patients with Cold-Agglutinin Hemolytic Disease (CAHD), and the hemorheological profile (deformability and osmotic fragility) by the utilization of rheo-optical techniques. The strength of the antigen-antibody reaction was approached by the work required to dissociate mechanically red blood cells agglutinates. It is focused on the evaluation of the qualitative adhesiveness of cell approached by the dissociation kinetics carried out in a Couette flow (erythroaggregameter). The analysis was performed by recording the increase of the reflectivity signal as the agglutinates are dissociated by shear into smaller ones. A total of eight patients aged <54 years with recent diagnostic of CAHD detected by positive Direct Anti-globulin Test (DAT) and very low RBC counts at 20 degrees C, were studied. Two parametric values were interesting: the dimensionless energy parameter and the characteristic dissociation time, which showed good correlation with hematological parameters. In conclusion, the dissociation method provides a powerful tool for estimating the qualitative adhesiveness of red blood cells agglutinated by autoantibodies in patients suffering of cold-agglutinin hemolytic disease and it would be very interesting to evaluate the severity of the disease.

Electrophoretic light scattering method has been considered to determine both the mean and polydispersity of electrophoretic mobility of normal human red blood cells. The final goal of our study was to optimize an in vitro test allowing us to investigate the interaction of xenobiotics, in particular polyelectrolytes, with blood cells. The feasibility of our method has been evaluated based on the reproducibility of our technique to analyze the native electrophoretic mobility of human RBCs, as well as their evolution in the presence of polycationic compounds, i.e. a natural polyamine, spermine, and a synthetic polycation, a polyethylenimine (PEI). Additionally, the follow-up of the human RBCs electrophoretic mobility has been controlled after their incubation with neuraminidase.

Hypertension (HTA) and dyslipidemia (DLP) represent major risk factors for cardiovascular and cerebral-vascular ischemic disease. The mechanisms through which they can induce vascular damage are both metabolic and mechanical. Hemorheological alterations in HTA are result of changes affecting both red cell intrinsic structure and their interactions with the plasmatic components. Several hemorheological determinants (biochemical, ionic, metabolic and rheologic) could influence and produce an impaired erythrocyte deformability determining an increased flow resistance in the microcirculation. The "Erythrodeformeter" allows obtaining the stationary and dynamical linear parameters of erythrocyte membrane by laser diffractometry. Stationary and oscillatory shear-induced elongation of cells leads to an elliptical diffraction pattern, its geometric characteristics being directly related to those of deformed RBC. Erythrocyte stationary parameters (Deformability Index, surface viscosity and elastic modulus) were obtained in stationary regime. Complex viscoelastic parameters (dynamic elasticity, dynamic loss, viscous and elastic components of the complex viscosity) were obtained when operating in oscillating mode. The diffractometric method is sensitive to detect pathological or induced alterations on RBC membrane, which can affect blood flow in vivo. The rheological parameters obtained give important information about the erythrocyte membrane and allow to detect and characterize erythrocyte alterations in vascular pathologies.

RBC flow cytometric analysis is usually used to quantify antigen content. Calibration systems enable antigen content determination by relating mean fluorescence intensity with the number of bound antibody molecules (equivalent to the number of antigen molecules). For that reason, antibodies must be used at saturating concentration, which may lead to agglutination when working with high density antigens. Then, forward scattering, side scattering and fluorescence will be increased, thus obtaining wrong results. In this work, the simple Langmuir adhesion model was applied. Flow cytometry was used to quantify GPA, a transmembrane protein present at high density on RBC. The fluorescence intensity of samples at different anti-GPA sub-saturating concentrations was measured. Sometimes, agglutinates were present and two peaks of fluorescence were observed, the principal one corresponding to isolated cells and the secondary one corresponding to agglutinated cells. In those cases, the principal peak was taken into account for the analysis. The GPA antigen content obtained for nine analyzed samples ranged from 3 to 13 x 10(5) sites per cell, which is similar to those values found in literature. Therefore, the Langmuir adsorption model enables us to determine the antigen content for the anti-GPA/GPA system on RBC membrane. This model could be used to quantify high density antigens in RBC and in other cells.

A higher than normal glucose concentration in a suspending medium may produce non-enzymatic glycosylation of erythrocyte proteins. This process can modify the viscoelastic properties of erythrocytes. In this paper, we studied the possible relationship between glucose concentration in a suspending medium and erythrocyte rheological parameters. Human venous blood was obtained from the antecubital veins of 10 healthy volunteers. Blood samples were anticoagulated with EDTA and centrifuged. Red blood cells (RBCs) were washed and subsequently divided in aliquots, which were incubated in vitro with glucose solutions of different concentrations. Dynamic and stationary viscoelastic parameters of RBCs were determined by laser diffractometry in an Erythrodeformeter. Aggregate shape parameter (ASP) of the RBCs was determined by digital image processing. Significant changes were observed both in ASP and in rheological parameters when the glucose concentration in the medium was increased, demonstrating that a glucose concentration as low as 1% induces alterations in the mechanical properties of RBCs.
